The project will investigate the role of the serine protease "fibroblast activation protein-alpha" (FAP) in facilitating metastasis by enabling breast cancer cells to thrive in the diverse microenvironments that they encounter as they leave the breast and colonize other organs. In particular, the potential production of pro-angiogenic and pro-invasive fragments of type I collagen by the proteolytic functions of FAP and matrix metalloproteinase-9 (MMP-9) will be investigated. In addition, the potential stabilization of metastatic microemboli by FAP proteolytic activity will also be explored. The successful fellow will become proficient in animal models of human breast cancer as well as in vivo and in vitro methods for determining tumor invasion and metastasis.
