FACSAria

To have cells sorted or analyzed on the FACSAria, contact the flow cytometry facility for an appointment and to confirm the protocol for sorting.

Due to software compatibility issues, the FACSAria is not on the UAMS network, and therefore cannot be used to deposit data on resnet.  Please bring a writeable CD, a USB portable storage drive, or a Zip disk to save your data

Samples must be provided in a 12 x 75 mm polystyrene tube (Fisher brand # 14-9563A or equivalent).  It is the responsibility of the investigator to make sure the cells are aggregate-free.   Samples can be filtered through a  strainer cap (35 micron, Falcon catalog number 35-2235) to remove aggregates.  This is extremely important since large clumps of cells can damage the flow cell tip, which is costly to replace and will cause flow cytometry facility downtime.  Filtration is absolutely required for samples to be run on the FACSAria .

Nozzles:  70 micron (up to 10 micron cells) and 100 micron (10-15 micron cells)

***Cells MUST be filtered before running on the FACSAria.  The nozzles are easily clogged by cell aggregates.***

Sample Acquisition Rate:  Maximum acquisition rate: 70,000 events per second
with 8 parameters, 12 compensation pairs.  Maximum rate < 70,000 events per second with more parameters, > 100,000 events per second with fewer parameters.

Sort Capability:  At 70 psi and 90 kHz (using the 70 micron nozzle) with an average threshold rate of 25,000 events per second for a four-way sort results in a purity of > 98% and a yield > 80% of Poisson's expected yield.

Automatic Cell Deposition Unit (ACDU) option allows for sorting into 6-, 24-, 48-, 96-, and 384-well plates or onto glass slides

Aerosol Management Option (AMO):  Equipped with a 0.01-µm size ultra-low penetrating air (ULPA) filter to trap aerosolized particles.  Evacuates the volume of the ACDU chamber area 16 times per minute in normal evacuation mode and 69 times per minute in rapid evacuation mode.
