Roles of mitochondria and mitochondrial DNA in apoptosis and cancer progression

Mitochondria are now considered a key organelle regulating apoptosis.
We showed that depletion of mitochondrial DNA (mtDNA) reduced apoptosis induced by specific mediators. Several characteristics associated with metastatic competence, such as invasion and overexpression of the tumor-specific regulator cathepsin L and transforming growth factor- can also be induced by depletion of mtDNA. Thus, mutation or loss of mtDNA could contribute to cancer development by inhibiting apoptosis or inducing metastatic competence. Certain cancers have a high frequency of large deletions in the mtDNA in addition to point mutants. Cells with a high proportion of large deletion mutant mtDNA lack some or most mtDNA-encoded proteins and therefore might show characteristics similar to those of cells with depleted mtDNA. Our studies are designed to investigate the mechanisms and the roles of accumulation of mutant mtDNA as a potential cause of cancer progression.
